根癌农杆菌介导花生高效遗传转化体系的优化

摘要

本文研究表面活性剂(MES,Methyl Ester Sulfonate))、真空渗入法和二次侵染对提高花生转化效率的影响,建立了一种农杆菌介导的花生快速高效遗传转化体系。该体系以带子叶的胚轴为外植体,在OD600为0.7的菌液中,加入150mg/L的MES,40kpa真空处理10min,共培养3d后进行二次侵染。采用该体系可显著提高遗传转化率,GUS基因阳性表达率最高,达73.3%。
